Abstract
YH1885 is a new candidate for therapy to stomach ulcer. The mutagenicity of YH1885 was studied by a micronucleus test in male ICR mice. The maximally tolerated dose(MTD) of YH1885 was determined to 5g/§¸. The doses adopted for the micronucleus test was therefore 5g/§¸ as a high dose, 2.5g/§¸ as s medium and 1.25g/§¸ as a low dose, respectively. Each of group was consisted of three doses of YH1885 solution, positive control 2§·/§¸ of mitomycin C and negative control 20 §¢/§¸ of 0.5% methylcellulose 1500. A slide preparation was made at 48 hours following administration.
No significant induction of microuclei was observed in any of the three doses of YH1885 administered per orally. No cytotoxicity such as inhibition of hemopoiesis was observed in any group of test agent as the rate of polychromatic erythrocytes to total erythrocytes was over 30%.
These results indicate that YH1885 is not capable of inducing micronuclei in in vivo mouse cells and thus has no genotoxicity in micronucleus test.
